Great job Miguel and Kevin! The chapter lays out the procedures of orthogonal/bioorthogonal intracellular labeling using the combination of SNAP/CLIP-tags and bioorthogonal chemistry (strain-promoted azide-alkyne cycloaddition and inverse-demand Diels-Alder reaction).

This work demonstrates the collective power of protein (SNAP/CLIP) tags and bioorthogonal reactions in intracellular dual-labeling experiments. Thanks to everyone who's involved in contributing to this work. The revision could not have been done on time if Miguel did not happen to be back in Tallahassee during the initial chaotic phase of the coronavirus pandemic.

It's been a while...

Since the last update, Miguel has graduated with his PhD, Peiye finally has the alkynyltriazole synthesis paper published, and the NSF has reaffirmed its continuous support to our research. Our group hosted Kristofer Kristofferson from Ohio Northern University as a REU student in the summer of 2019. Also, our REU alumnus Brandon has returned as a graduate student, while Kevin joined our group as part of Class 2024. We also welcomed Justin Lee, Dr. Manoj Manna, Dan Stone, and Jared Cossin as new additions. So it has been a while!

Miguel expanded the substrate scope of SNAP-tag, an important tool in bioconjugate chemistry. In addition to benzylguanine-type substrates, 5-pyridylmethylguanine (5PG) derivatives are also taken up by SNAP-tag with comparable efficiencies.
